Product Overview
DSIP (Delta Sleep-Inducing Peptide) is an endogenous nonapeptide with the amino acid sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, also known by its INN name Emideltide. It was first isolated in 1977 by Monnier and colleagues from the extracorporeal dialysate of cerebral venous blood in rabbits subjected to hypnogenic electrical stimulation of the intralaminar thalamic area — an elegant cross-perfusion experiment that launched the modern era of sleep-factor research.
When infused into the mesodiencephalic ventricle of recipient rabbits, DSIP induces spindle and delta EEG activity — the electrophysiological signature of slow-wave (deep) sleep. Subsequent work detected DSIP-like immunoreactivity in neurons, peripheral organs, cerebrospinal fluid and plasma, suggesting roles beyond sleep itself: published research has explored DSIP in stress-response modulation, neuroprotection after cerebral ischemia, and seizure models.
Each vial contains 5mg of high-purity DSIP as a white to off-white lyophilized powder, supplied for in-vitro and laboratory research use only.

🐇 1977, Basel — Monnier's laboratory applied low-frequency electrical stimulation to the intralaminar thalamic area of "donor" rabbits to induce slow-wave sleep, then collected the cerebral venous blood via extracorporeal dialysis.
🔁 Transfer to recipients — the dialysate was infused into "recipient" rabbits, which showed increased spindle and delta EEG activity even without stimulation: a humoral sleep factor had been transferred. Amino-acid analysis and sequence determination identified the active component as the nonapeptide Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, and chemical synthesis confirmed the activity (Schoenenberger & Monnier, 1977).
🌍 An unresolved riddle — decades later, DSIP's natural occurrence and primary physiological role remain actively debated in the literature (Kovalzon, 2006), which is part of what keeps DSIP a compelling research subject.
Mechanism & Researched Pathways
🌙 Slow-wave sleep promotion — central administration induces spindle and delta EEG activity in animal models; the signature effect that named the peptide. 🧠 Stress-response modulation — studied for effects on stress-induced physiological changes and corticosteroid regulation in animal models. 🛡 Neuroprotection research — reported to reduce neuronal activity changes and improve blood supply in brain-ischemia models; associated with motor-function recovery after ischemic challenge (Tukhovskaya et al., 2021). ⚡ Excitability modulation — evaluated in generalized-seizure models for modulatory effects on excitatory/inhibitory balance. 🔍 Systemic distribution — DSIP-like immunoreactivity detected in brain, cerebrospinal fluid, plasma and peripheral tissues, supporting investigation of extra-sleep functions. Research Highlights
📖 Schoenenberger & Monnier, 1977 — Isolation & Sequence
The defining study: isolation from rabbit cerebral venous dialysate, amino-acid analysis, sequence determination and synthesis of the sleep-active nonapeptide.
📖 Tukhovskaya et al., 2021 (Molecules)
Delta sleep-inducing peptide recovered motor function in a brain-ischemia model, adding neuroprotection to DSIP's documented activity profile.
📖 Kovalzon, 2006 — The Unresolved Riddle
A widely cited review summarizing the open questions around DSIP's natural occurrence and biological activity — essential reading for researchers designing DSIP studies.
📋 Evidence Status Note
DSIP's sleep-inducing effects are primarily documented in animal models (rabbit intraventricular infusion) and a small number of early human studies with mixed outcomes. We present this evidence hierarchy transparently so researchers can evaluate the compound appropriately for their own study designs.
Research Applications
🌙 Sleep-neuroscience — slow-wave sleep factor and EEG-activity studies (institutionally approved protocols only). 🧪 Stress-physiology research — neuroendocrine and behavioral stress-response readouts. 🧬 Neuroprotection assays — ischemia models, neuronal-survival and motor-recovery endpoints. 📊 Immunoassay / quantitation work — DSIP-like immunoreactivity detection and distribution studies. Reconstitution & Handling
💧 Add sterile or analytical-grade water slowly down the side of the vial; swirl gently until fully dissolved — do not shake vigorously. 🧪 Aliquot reconstituted solution to avoid repeated freeze-thaw cycles; label each aliquot with batch, concentration and preparation date. 🧊 Lyophilized powder: store frozen (-20°C), protected from light and moisture; reconstituted solution: refrigerate and use within your validated stability window. ⚖️ Handle with aseptic technique and appropriate PPE per your laboratory SOP. Quality Assurance
✔ Lot-specific Certificate of Analysis: HPLC purity confirmation + mass-spectrometry identity verification (expected [M+H]⁺ consistent with the nonapeptide mass of 848.8).
✔ Third-party batch testing documentation available upon request.
