AICAR 50MG 100MG

The peptide will be provided as lyophilized powder to ensure maximum stability.
AICAR is a cell‑permeable purine nucleoside analog, a well‑known pharmacological AMPK activator for laboratory research only, not a peptide.
Inside cells, AICAR is converted into ZMP, an AMP mimic that allosterically activates AMP‑activated protein kinase (AMPK), the master regulator of cellular energy homeostasis. In pre‑clinical studies, AICAR is widely applied to explore glucose uptake, fatty‑acid oxidation, mitochondrial biogenesis, lipid metabolism and inflammatory response. It is also investigated as an “exercise‑mimetic” compound in animal models for endurance‑related metabolic adaptations.
Our AICAR lyophilized powder is tested by HPLC‑MS, each batch comes with complete COA documentation.

    Product Overview

AICAR (5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside), also known as Acadesine or AICA riboside, is a cell-permeable adenosine analog and the classical direct activator of AMP-activated protein kinase (AMPK) — the master kinase of cellular energy homeostasis. Note: AICAR is a small-molecule nucleoside, not a peptide.

Once taken up by cells through adenosine transporters, AICAR is phosphorylated to ZMP (AICA ribotide), which mimics AMP: it binds the same site on the γ subunit of AMPK, allosterically activates the enzyme, and stimulates phosphorylation at Thr172 by the upstream kinase LKB1 — all without changing cellular ATP, ADP or AMP concentrations. This mechanism distinguishes AICAR from indirect activators such as metformin, which acts by creating genuine energetic stress at mitochondrial Complex I.

In cellular and animal studies, AMPK activation by AICAR promotes insulin-independent glucose uptake in skeletal muscle, shifts lipid metabolism toward fatty-acid oxidation, and remodels adipocyte metabolism toward energy dissipation. Beyond metabolism, acadesine has a long research history in cardiac ischemia and reperfusion protection. The compound has also been widely described in the scientific literature as a candidate "exercise mimetic" — a molecule that activates exercise-like metabolic signaling pathways.

Each vial contains 100mg of high-purity AICAR as a white to off-white crystalline powder, supplied for research use only.

    Mechanism of Action

    Step What Happens
    1. Cellular Uptake AICAR enters cells through adenosine transporters — no transfection or special delivery required.
    2. Phosphorylation to ZMP Intracellular kinases convert AICAR to ZMP (AICA ribotide, C9H15N4O8P), the true AMP-mimetic effector.
    3. AMP-Site Binding ZMP binds the AMP-binding site on the γ subunit of AMPK, producing allosteric activation — while cellular ATP, ADP and AMP levels remain unchanged.
    4. Thr172 Phosphorylation Binding stimulates phosphorylation at Thr172 by the upstream kinase LKB1, fully activating AMPK kinase activity.
    5. Downstream Signaling Activated AMPK promotes catabolic energy-producing pathways: insulin-independent glucose uptake in muscle, fatty-acid oxidation, mitochondrial biogenesis, and suppression of anabolic processes.

AICAR vs Metformin: Two Routes to AMPK

    Aspect AICAR Metformin
Activation Mode Direct — AMP-site mimicry via ZMP Indirect — inhibits mitochondrial Complex I, creating genuine energetic stress
Nucleotide Levels ATP / ADP / AMP unchanged AMP/ATP ratio rises due to energetic stress
Research Value Clean tool for isolating AMPK-specific signaling effects Studies combining energetic stress with AMPK activation

Mechanistic comparison summarized from the published AMPK-activator literature; both compounds ultimately converge on AMPK activation.

    Research Highlights

📖 Kim et al., 2016 (Experimental & Molecular Medicine)

Defined AICAR as the first direct AMPK activator — an adenosine analog taken up into cells and converted to the AMP-mimetic ZMP. The review remains a standard reference for AMPK-activator mechanisms.

📖 Gaidhu et al., 2009 (Journal of Lipid Research)

Provided evidence that prolonged AICAR-induced AMPK activation can remodel adipocyte metabolism by upregulating pathways that favor energy dissipation — a key finding for fat-metabolism research.

📖 "Exercise Pills: At the Starting Line" (Trends in Pharmacological Sciences, 2015)

Positioned AICAR among the prototype exercise mimetics — compounds that activate the same AMPK-driven metabolic pathways triggered by muscle contraction.

📖 Višnjić et al., 2021 (systematic review, 180+ citations)

A systematic review of AICAr as a widely used AMPK activator — confirming its activation mechanism (γ-subunit binding, Thr172 phosphorylation via LKB1) while also cataloguing important AMPK-independent effects to consider in experimental design.
